EOS plays an important role in maintaining the suppressive function of regulatory T cells (Treg), and a set of Tregs induced transformation into T helper-like cells, which are able to secrete proinflammatory cytokines in response to certain inflammatory signals. Meanwhile, a significant reduction in Treg activity along with the production of proinflammatory cytokines have been reported in patients with HTLV-1-associated myelopathy / tropical spastic paraparesis (HAM / TSP) .In this study, to test whether there is a change in the expression of EOS in mononuclear cells peripheral blood (PBMC) were from individuals HTLV-1-infected mainly HAM / TSP, we examined the expression of HTLV-1 genotype tax proviral load (PVL), and the mRNA expression of taxes, HBZ and EOS in HTLV- 1 infected individuals, including cell T adult leukemia / lymphoma (ATL), HAM / TSP, or asymptomatic carriers.
EOS expression levels of mRNA and protein in various HTLV-1-infected or uninfected human T cell line investigated.EOS also highly expressed at the protein level in most of HTLV-1 line-infected T cells, and added after the HTLV-1 Tax regulatory factors induced in the JPX-9 Tax-induced cell lines. Immunoprecipitation experiments showed physical interaction between EOS and Tax viral regulatory protein, but not HBZ. Meanwhile, there was a significant reduction in the level of mRNA in PBMCs EOS HTLV-1 infected individuals regardless of their clinical status.
We found an inverse correlation between the level of the EOS mRNA and HTLV-1 PVL in patients with ATL, and a positive correlation between the two load EOS mRNA and PVL, and EOS and load HBZ mRNA in patients with HAM / TSP, whereas this correlation was not observed in other clinical findings statuses .
These show that both Tax and HBZ can alter the expression of EOS through a mechanism that has not been determined, and dysregulation EOS expression in PBMCs HTLV-1 infected individuals may contribute to the pathological development of HTLV-1-related diseases, such as ATL and HAM / TSP.

The possible involvement of microRNAs (miRNAs) in malignant pleural fluid were investigated using small RNA sequencing. regulatory T cells (Treg) markers (CD4, CD25, forkhead box P3) and Helios (also known as IKAROS Family Zinc Finger 2 [IKZF2]) were detected using flow cytometry. The expression level of miR-4772-3p IKZF2 and measured using real-time quantitative reverse transcription polymerase chain reaction. Interaction between miR-4772-3p and Helios determined using dual-luciferase reporter test.
Effects of miR-4772-3p on Helios expression was evaluated using in vitro system. test the correlation between miR-4772-3p and functional molecules Tregs that performed.Compared with non-malignant controls, patients with lung cancer non-small cell has an increased frequency of Tregs with MPE and Helios expression in peripheral blood mononuclear cells. the downregulation of miR-4772-3p verified inversely proportional to the frequency of Tregs Helios and Helios expression in the MPE. Overexpression of miR-4772-3p could inhibit the expression of Helios in vitro experiments.
